Aging isn’t inevitable—at least not in the way most people think.

Dr. David Sinclair, one of the world’s leading scientists in aging research, has spent three decades proving that how we age is largely within our control.

In a recent episode of A Life of Greatness podcast, Sinclair revealed groundbreaking discoveries about reversing aging at the cellular level—and how close we are to testing these technologies in humans.

What he shared could fundamentally change how we think about disease, longevity, and what it means to grow old.

The Information Theory of Aging: Your Body as a Computer

Sinclair’s research challenges the traditional “wear and tear” model of aging. Instead, he proposes something far more hopeful: the information theory of aging.

Our bodies are much more like computers where the software gets corrupted over time. This is the biggest thing that we’re finding in the lab that there’s a backup copy of that software in every cell that we have in our bodies that allows us to reinstall it and every cell in the body can be made to be young again.

According to Sinclair, diseases like cancer, heart disease, Alzheimer’s, and diabetes aren’t separate conditions—they’re all manifestations of one universal process: loss of cellular information.

FULL-BODY TRAINING

If you want something simple that actually works, this is one of the most effective tools I’ve used to build strength, conditioning, and endurance without needing a full gym setup.

  • Full-body training with one weight using swings, squats, and presses
  • Solid cast iron build that feels stable and lasts for years
  • Comfortable grip that makes high-rep workouts easier to handle

That information exists in two forms: the genome (your DNA) and the epigenome (the control systems that read your DNA). While DNA damage plays a role, Sinclair believes epigenetic disruption is the real driver of aging.

The revolutionary part? There appears to be a backup copy of youthful information in every cell—and scientists are learning how to restore it.

Reversing Blindness in Mice—And Humans Are Next

In 2020, Sinclair’s lab published proof that they could reverse aging in animals.

We chose a very difficult thing to cure which was blindness. And we literally reversed the age of the retina and caused mice to be able to see.

The technology, called ER-100, works by resetting the epigenetic markers in cells—essentially reinstalling youthful software.

When asked how far away human trials were, Sinclair’s answer was stunning:

About a week.

The FDA approved trials in January 2026. Sinclair expects to meet the first patient soon—a person who will receive a single injection in the eye and potentially regain vision within weeks.

The technology has already shown success in non-human primates, whose eyes are nearly identical to humans. It’s also worked in other parts of mouse bodies: brain, kidneys, liver.

Age reversal in humans is no longer science fiction—it’s happening now.

What Accelerates Aging (And What Slows It Down)

Sinclair emphasized that more than 80% of aging is determined by lifestyle, not genetics.

Everybody listening has to know aging is malleable. Some estimates are 80% plus. It depends on how you live your life.

What Speeds Up Aging

  • Sunburn – UV damage corrupts cellular information
  • Excessive alcohol – Particularly damaging over time
  • Smoking – Sinclair calls it “probably the worst thing you can do”
  • Sitting all day – Lack of movement signals abundance mode
  • Overeating – Constant eating turns off longevity genes
  • Chronic stress – High cortisol accelerates cellular aging
  • Loneliness – Measurable acceleration of biological age

You can see lonely people, they do have accelerated aging. If you’re lonely and you don’t have a partner, find one. If you can’t find a partner, get a pet.

What Slows Aging Down

Sinclair advocates for mild adversity—the Goldilocks zone between comfort and crisis.

  • Intermittent fasting – Not eating constantly activates longevity pathways
  • Regular exercise – Especially strength training and walking
  • Quality sleep – Critical for cellular repair
  • Strong social connections – Real friends, not hundreds of acquaintances
  • Limited sun exposure – Gradual, protected exposure without burning
  • Plant-focused diet – Preferably with minimal or no red meat

Sinclair himself eats one to one-and-a-half meals per day, maintains 9-12% body fat, and works out daily while watching TV or listening to podcasts.

His 86-Year-Old Father: The Ultimate Case Study

When Sinclair talks about successful aging, he points to his father as living proof.

My father is my hero when it comes to health. He’s 86 turning 87 in a few months and there aren’t that many 87 year old men who are in perfect health let alone still alive.

His father eats one meal per day, walks six kilometers regularly, goes to the gym three times weekly, and is stronger and fitter than Sinclair himself.

The regimen both father and son follow includes:

  • NMN – An NAD booster Sinclair has taken for 15 years
  • Metformin – A diabetes drug that regulates blood sugar
  • Resveratrol – Activates sirtuin longevity enzymes
  • Fish oil – For cardiovascular and brain health
  • Baby aspirin – Anti-inflammatory benefits
  • Nattokinase – May reduce arterial plaque over time
  • Alpha lipoic acid – Supports metabolism

Sinclair is careful to note these aren’t medical recommendations—just what works for his family after decades of self-experimentation and monitoring.

Cancer as an Age-Related Disease

Sinclair’s view of cancer is unconventional but compelling.

While most researchers focus on DNA mutations, Sinclair argues that epigenetic dysfunction is equally critical. Young cells can harbor cancer-causing mutations without developing tumors because their epigenome maintains proper control.

You and I have cancer cells in our body right now, but they don’t grow because the cell has the epigenome. And the immune system is still young and can kill those cancer cells.

His student has shown that reversing the age of cancer cells can slow or even kill them—the cells essentially realize they don’t belong and self-destruct.

This approach could revolutionize cancer treatment by addressing the root cause rather than just targeting mutations.

The Power of Social Connection

Sinclair’s perspective on relationships has evolved dramatically with age.

I used to have hundreds of friends. Now my group is probably four people. My three kids and my partner and I call my dad every day in Australia and that’s it. That makes me the happiest.

Research from Harvard’s Robert Waldinger confirms what Sinclair has learned personally: quality relationships are among the strongest predictors of longevity.

Loneliness doesn’t just feel bad—it’s measurable in blood tests as accelerated biological aging.

During the interview, Sinclair received a text from his 22-year-old daughter saying she missed him—a moment he described as more meaningful than any scientific award.

Beyond Blindness: What’s Coming Next

Sinclair’s lab isn’t stopping with vision restoration. They’re testing age reversal technology on:

  • Menopause – Developing oral treatments to delay or prevent it
  • Skin aging – Testing wound healing and hair regrowth
  • Cognitive decline – Brain rejuvenation studies ongoing
  • General frailty – Muscle, liver, and kidney restoration

Sinclair is also using AI to accelerate research and reduce costs, potentially making treatments accessible to millions rather than just the wealthy.

The Mission Behind the Science

Sinclair’s grandmother Vera shaped his life’s work with a blunt statement when he was four years old:

Everyone dies eventually and I’m going to die, your parents are going to die, your cat is going to die, and you’re going to die and then we’re all gone forever.

That moment never left him. Neither did watching his mother suffer for 20 years with one lung after cancer surgery, eventually dying at 70.

In contrast, his father at 86 contributes to society, mentors his grandchildren, and loves life—starting new careers and helping his community.

I want that life for everybody. I don’t worry about living forever. That’s crazy talk. Let’s focus on getting it right here, right now.

When criticized for working on longevity, Sinclair’s response is fierce:

What kind of a cruel human being are you? Have you never been to a nursing home watching people having to live the same 5 minutes over and over again? I am going to work hard every day with my team to try and overcome that suffering for as many people.

A New Model of Healthcare

Sinclair is building lifespan.com—a global community focused on evidence-based longevity science. The platform will include:

His vision echoes what he’s seen work at companies like SpaceX—when humans pull in the same direction without politics or fighting, extraordinary things become possible.

Within a decade, Sinclair predicts doctors will prescribe age-reversal treatments—not just for specific diseases, but for aging itself.

The Best Advice He Ever Received

From genomics pioneer George Church, Sinclair learned a career-defining lesson:

Just ignore everybody and do what you want.

Five years ago, scientists vilified Sinclair for using the term “age reversal.” Now it’s the hottest area in biology.

He was attacked for speaking directly to the public rather than staying in his lab. Now millions have changed their lives based on his work.

Ignoring critics and following his calling has defined Sinclair’s greatest contributions.

What Defines a Life of Greatness

For Sinclair, greatness isn’t about fame or achievement—it’s about purpose.

A life of greatness is living the best possible life you can live and making other people’s lives the best lives they can also live and leaving the world of course a better place than you found it.

He believes humans give the universe consciousness and meaning—that we’re here to protect this “treasure called planet Earth” and spread awareness throughout the cosmos.

Whether through evolution or something higher, humanity has a responsibility to live nobly and help others do the same.

With age reversal trials beginning in humans, Sinclair’s three-decade mission is entering its most exciting phase—one that could fundamentally transform what it means to grow old.
