The Digestion Argument Has Been Debunked

Dr. Norton’s initial skepticism centers on basic digestion.

If you eat collagen, if you ingest a supplement, that supplement gets broken down during digestion to the constitutive amino acids, it’s not like you take in collagen and it just goes right into your connective tissue.

He’s absolutely right that collagen breaks down in the stomach. Hydrochloric acid unfolds protein structures, while enzymes like pepsin chop them into smaller pieces.

However, this argument misses critical nuance. Research shows collagen doesn’t just become individual amino acids.

Studies demonstrate that di-peptides and tri-peptides (two or three amino acids still bound together) appear in human blood after collagen consumption. These small peptide chains are bioactive, meaning they trigger cellular responses.

When scientists apply collagen peptides directly to skin cells in laboratory studies, those cells respond by producing more structural proteins. The peptides essentially signal cells to ramp up collagen production, rather than simply providing raw materials.

The Meta-Analysis That Started Everything

A recent meta-analysis examined collagen supplementation’s effects on skin elasticity, hydration, and wrinkles. Initial results looked promising across all three measures.

But Dr. Norton identified a massive red flag in the sensitivity analysis.

In the sensitivity analysis excluding outlier studies showing an extreme beneficial effect, collagen supplements showed no significant improvement of wrinkles and a decreased effect for skin hydration and elasticity.

One study showed such dramatic benefits that when researchers removed it, results either disappeared or drastically weakened. That’s concerning because single studies shouldn’t dominate entire meta-analyses.

Still, even after removing outliers, two out of three skin measures showed some positive effect.

Studies that did not receive funding from pharmaceutical companies showed no significant effect of collagen supplements for improving skin hydration, elasticity, and wrinkles, while those funded showed significant improvements in all categories.

Industry-funded studies also scored lower on methodological quality. High-quality studies revealed no significant benefits, while low-quality studies did.

This appears damning. When independent researchers conduct rigorous studies, collagen benefits vanish.

The Plot Twist Nobody Expected

Examining those seven independent studies individually reveals something shocking.

Six out of seven independent studies actually found collagen supplements improved skin.

Only one showed no effect. Dr. Stanfield noticed the same pattern with high-quality studiesโ€”seven out of nine demonstrated positive impacts from collagen peptides.

How can individual studies show benefits while the combined analysis concludes otherwise?

Statistical Constraints Muddy Waters

Multiple factors explain this apparent contradiction.

First, segmenting 23 studies into subgroups drastically reduces available data. Dividing by funding source, study quality, and specific skin metrics sometimes left researchers analyzing just one to three studies.

With such limited data, statistical confidence weakens dramatically. Results that might indicate real effects get classified as “no significant effect” due to insufficient power.

Additionally, high-quality study subgroup analyses with 11-12 studies did identify positive effects, supporting the idea that more data reveals clearer patterns.

Valid Critiques Remain

Dr. Norton raises legitimate concerns beyond funding bias.

No collagen skin studies compare supplementation against regular protein. All compare against placebo, making it impossible to determine whether collagen peptides offer unique benefits versus simply consuming more protein or amino acids like glycine and proline.

One burn healing study suggests collagen outperforms standard protein, but more evidence is needed.

Another consideration: realistic expectations matter. Nobody claims collagen peptides reverse decades of aging overnight.

If effects are modest rather than dramatic, detecting them requires larger, higher-quality studies. Current research may simply lack sufficient power to capture small-to-moderate improvements consistently.

What Should Consumers Believe?

The evidence presents a nuanced picture rather than clear-cut answers.

Science rarely provides absolute certainty, especially for interventions with subtle effects. Collagen supplementation falls squarely into this gray areaโ€”promising enough to justify cautious optimism, but requiring better research before declaring victory.
